Description:
T. brucei encodes two phosphatidylinositol 4-kinases. Phosphatidylinositol 4-kinase Beta (TbPI4KIII-ß), has been demonstrated to be essential in procyclic form T. brucei by RNAi (1). The protein has been expressed in COS-7 cells, and its activity confirmed. Depletion of TbPI4KIII-ß by RNAi arrested cell growth and distorted the cellular morphology. The Golgi apparatus was distorted and lysosomal and flagellar pocket proteins were mislocalized. A twisted phenotype was observed by scanning electron microcopy. Dividing cells exhibited a detached daughter flagellum and lacked a cleavage furrow. Cell cycle analysis showed that depletion of TbPI4KIII-ß caused a post-mitotic cytokinesis block that occurs after a one round of mitosis. 1. M. J. Rodgers, J. P. Albanesi, M. A.
